Turn that around a little and you can do it now. Create a profile called 'Template' (etc), and set everything as you want it (including Shared Settings, if you want to use them), but with a dummy Source & Destination.

When you want a new profile, just Copy that Template profile to a working name, open it for Modification, then drag & drop the Source & Destination you want into the boxes (or, edit them manually if you prefer). Close profile - done...

You can save several of these templates, with different configurations (Fast Backup, etc) and name them accordingly.
